Subcellular localization of Forssman glycolipid in epithelial MDCK cells by immuno-electronmicroscopy after freeze-substitution
Forssman antigen, a neutral glycosphingolipid carrying five monosaccharides, was localized in epithelial MDCK cells by the immunogold technique. Labeling with a well defined mAb and protein A-gold after freeze-substitution and low temperature embedding in Lowicryl HM20 of aldehyde-fixed and cryoprotected cells, resulted in high levels of specific labeling and excellent retention of cellular ult...
متن کاملCapping of exogenous Forssman glycolipid on cells
When motile cells are incubated with Forssman glycolipid, the antigen is incorporated into the cells' plasma membranes. If cross-linked by antibody, the patched glycolipids cap. This process is sensitive to those drugs that are known to inhibit capping of protein antigens. The results support a flow mechanism for capping.
متن کاملMolecular genetic basis of the human Forssman glycolipid antigen negativity
Forssman heterophilic glycolipid antigen has structural similarity to the histo-blood group A antigen, and the GBGT1 gene encoding the Forssman glycolipid synthetase (FS) is evolutionarily related to the ABO gene. The antigen is present in various species, but not in others including humans. We have elucidated the molecular genetic basis of the Forssman antigen negativity in humans. In the huma...
متن کاملSubcellular localization of boron in cultured melanoma cells by electron energy-loss spectroscopy of freeze-dried cryosections.
Boron neutron capture therapy (BNCT) is based on the ability of the non-radioactive isotope 10B to capture thermal neutrons and to disintegrate instantaneously. This reaction opens a way to selectively destroy tumour cells after specific uptake of 10B. In this paper, a method based on electron energy-loss spectroscopy is presented for detecting and quantifying boron in freeze-dried cryosections...
متن کاملA revised structure for the Forssman glycolipid hapten.
A glycolipid hapten of the Forssman antigen of horse spleen was found to be a ceramide pentasaccharide, which contained 2 moles of N-acetylgalactosamine, 2 moles of galactose, and 1 mole of glucose per ceramide. An ar-N-acetylgalactosaminidase of hog liver (WEISSMAN, B., AND HINRICHSEN, D. F., Biochemistry, 8, 2034 (1969)) hydrolyzed the terminal a-N-acetylgalactosaminosyl residue with a simult...
